Extreme ketogenic, but not moderate, low-carbohydrate/high fat diets lead to loss of lean body mass in rats due to impairments of the GH/IGF system

Bielohuby M , Sawitzky M , Stoehr B , Ramisch J , Menhofer D , Hoeflich A , Bidlingmaier M

In rats, we have previously demonstrated that consumption of low-carbohydrate/high-fat diets (LC–HFD) led to significant reductions in circulating IGF1. We further observed that rats fed an extreme, ketogenic, but comparably low protein LC–HFD (LC–HF-2) showed loss of bodyweight, which was mainly due to loss of lean body mass (LBM). Consumption of a moderate protein LC–HF diet (LC–HF-1) did not affect LBM acquisition compared to controls. We now invest...

Investigating the effects of 5.5 mmoL vs 25 mmoL glucose concentration in culture media on LHCN-M2 cell viability, proliferation, metabolism and differentiation

Brett Ryan , Renshaw Derek , Hodson Leanne , Turner Mark

Introduction: In vitro skeletal muscle cell models are vitally important for investigating the molecular mechanisms of skeletal muscle in metabolic and endocrine diseases, such as obesity and type 2 diabetes. Culture media for skeletal muscle cells can often contain glucose concentrations (GC) five times higher than what’s considered normal in fasting human plasma, thus is not representative of the in vivo environment. Hyperglycaemia in culture media may...

Topical application of CD362+ human mesenchymal stem cells (cyndacel-M) seeded in Excellagen scaffold augments wound healing and increases angiogenesis in a diabetic rabbit ulcer model

Patil Swapnil , Chen Xizhe , Watson Luke , Loftus Paul , Flynn Lisa O' , Chandler Lois , Rubanyi Gabor , Elliman Stephen , O'Brien Timothy

Non-healing foot ulcers are a major complication in diabetic patients. Mesenchymal stem cells (MSCs) are known to promote angiogenesis with improved wound healing. Orbsen Therapeutics has identified a novel antibody (CD362+) which can be used to prospectively FACS-isolate CD362+CD45− MSC from human bone marrow with enhanced MSC/MNC purity ratios of up to 1/4. Excellagen matrix was used to seed the cells as biomaterials are well reported t...

Establishing an anti-müllerian hormone (AMH) cut-off to determine polycystic ovarian morphology (PCOM) supporting diagnosis of polycystic ovarian syndrome (PCOS): The APHRODITE study

Dietz de Loos Alexandra , Hund Martin , Buck Katharina , Meun Cindy , Sillman Johanna , Laven Joop

PCOS is a common pathology in women of reproductive age, and ~70% of affected women remain undiagnosed. Clinical features of PCOS include ovulatory dysfunction, hyperandrogenism and PCOM (Rotterdam criteria). Replacement of transvaginal ultrasound (TVUS) with a blood test for PCOM is clinically desirable, as lack of access to TVUS may contribute to underdiagnosis of PCOS. We aimed to derive and validate a cut-off for AMH to discriminate PCOM using the Elecsys AMH Plus imm...

Response of human adrenocortical carcinoma cell lines to cytostatic drugs and mitotane and the role of the multidrug resistance pump MDR1

van Koetsveld P M , Dogan F , Misier M B , Feelders R A , de Herder W W , Hofland L J

Introduction: In case of inoperable disease or tumor recurrence, therapy with mitotane (MIT), sometimes in combination with doxorubicin (DOX), etoposide (ETO), cisplatin (CIS) or streptozocin (STREP), is frequently applied in patients with adrenocortical carcinoma (ACC). The multidrug resistance pump MDR1 is responsible for resistance of tumor cells to cytostatic drugs. The activity of MDR1 is inhibited by the calcium-channel blocker verapamil. There are equivocal data in the ...

TCF7L2 gene variants and plasma C-peptide concentrations in diabetes type 2

Ferreira M C , Marques M C A , Fukui R T , Correia M R , Silva M E R , Rocha D M , Santos R F

Aims/hypothesis: Several data have shown the association between diabetes type 2 (DM2) and transcription factor 7-like 2 (TCF7L2) gene polymorphisms, in several and different ethnical populations. Although the pathophysiology is not completely clear, it seems that beta and alpha cell functions are abnormal. Up to these days there is no clinical data showing association between TCF7L2 gene polymorphism allele rs7903146 T and beta cell insulin and C-peptide contents in DM2 patie...

IGF acts through the IGF1R mediated PI3K pathway in first trimester human placenta to rescue cytotrophoblasts from apoptosis

Forbes K , Aplin JD , Westwood M

Conditions such as pre-eclampsia and intrauterine growth restriction highlight the importance of normal placental cell turnover for successful pregnancy outcome. In these conditions abnormal levels of proliferation and increased apoptosis have been reported; aberrations in components of the IGF axis have also been observed. IGFs regulate proliferation and survival in other cellular systems but their ability to influence human placental cell function remains to be established.<...

Acoustic radiation force impulse elastography (ARFI) in patients with autoimmune thyroid disease

Vlad M , Zosin I , Balas M , Sporea I , Bota S , Popescu A , Popa A , Radu S

Introduction: Elastography is a new dynamic technique that uses ultrasonography for the assessment of tissue stiffness. The principle of ultrasound-elastography is that compression of the examined tissue induces a smaller strain in hard tissues than in soft ones. Until now elastography has been applied to study the hardness and elasticity of thyroid nodules, aiming to differentiate malignant from benign ones.Aim: We decided to study if ultrasound elastog...

Vitamin D modulates composition of extracellular matrix in cultured human vascular cells

Ivanov V. , Ivanova S. , Niedzwiecki A. , Rath M.

Vitamin D (VitD) was shown to affect biology of vascular wall by regulating cell proliferation. However, its effects on vascular extracellular matrix (ECM) remain largely unaddressed. We investigated effects of VitD (calcitriol) on composition of ECM deposed by cultured endothelial cells (HAEC) and smooth muscle cells (HASMC) isolated from human aorta, and compared them to effects of ascorbate (VitC). Cellular monolayers were treated with or without vitamins for 72 h, then ECM...

Proliferative and apoptotic effects of bisphenol A and its combination with estradiol in human breast carcinoma cells

Havranek T. , Macho L. , Fickova M.

Bisphenol A (BPA) is an endocrine disruptor (ED) used as plasticiser present in plastic products of daily use. The leakage of BPA monomer into food, drinks represents the ubiquitous exposure to the general population. BPA stimulatory effect on breast cancer cells growth in vitro and increased incidence of hormone dependent malignancies suspected BPA from carcinogenic effect. The aim of this study was to investigate the effects of BPA alone and in t...
